Bioencapsulation of the antibacterial drug sarafloxacin in nauplii of the brine shrimp <i>Artemia franciscana</i> OMA

Nauplii of the brine shrimp <i>Artemia franciscana</i> were enriched with the antibacterial drug sarafloxacin hydrochloride to determine if levels effective against four pathogenetic strains of <i>Vibrio</i> spp. would accumulate in brine shrimp. Three vibrio strains were <i>Vibrio anguillarum</i>; the fourth was <i>V. vulnificus</i>. Concentrations of sarafloxacin ranging from 1 to 40% (weight: volume) were incorporated into the oil phase of Super Selco brine shrimp diets. Enriched nauplii and controls were sampled at 2h interval for 24h. Antibiotic sensitivity testing was evaluated with a modification of the Kirby-Bauer disk diffusion method. Effect of salinity on the uptake of cadmium by the brine shrimp <i>Artemia fransiscana</i> OMA

The effect of salinity on cadmium uptake by the brine shrimp <i>Artemia fransiscana</i> was studied in chemically defined saltwater solutions. Shrimp were acclimated to different salinities and exposed to the metal in solutions of differing salinity and composition. Within each acclimation group, the cadmium uptake decreased with increasing salinity of exposure. Most of this variation was explained by changes in the activity of the free metal ion. Among the acclimation groups, uptake increased by increasing salinity of acclimation. Most of this variation was explained by changes in the permeability of the shrimp. Effect of temperature on the uptake of copper by the brine shrimp, <i>Artemia franciscana</i> OMA

The effect of temperature on the uptake of copper by the brine shrimp <i>Artemia franciscana</i> has been studied in chemically defined saltwater solutions. Animals were acclimated to temperatures ranging from 10°C to 35°C and exposed to copper at different temperatures within this range. The results show that within each temperature acclimation group, copper uptake increases with increasing temperature of exposure. Within each temperature acclimation group most of the variation can be related to the combined effect of temperature on the chemical speciation and diffusion rate of copper in the diffusion layer lining the exchange surfaces. Total length and biomass production of the brine shrimp Artemia franciscana were studied fed on soybean and wheat micropulverized meals (applied alone or mixed at different proportions), live microalgae (Tetraselmis suecica and Chaetoceros calcitrans), and dried Spirulina as diets. Eight diets were tested in triplicates during 10 days. Significant differences (P < 0.05) were observed from day 1 onwards. The mixed meal-based diets showed better production results. At day 1, the Artemia nauplii fed on the 70% wheat meal/ 30% soya meal diet were 30% longer compared to the animals from the C. calcitrans group. We present evidence that incubation of brine shrimp <i>Artemia</i> encysted embryos (cysts) at high temperatures (36-37°C) suppresses hatching of cysts from a parthenogenetic population, while at the same time allowing cysts of the New World bisexual species (<i>A. fransiscana</i>) to hatch. Ultrastructural changes of the chorion were studied in prehydrated Artemia cysts. After incubation for 12, 24, 48 and 72 hours, in hygrostats (under controlled relative humidity, that range from 19% to 99%), the authors observed in cysts that were maintained (for at least 48 h in 99% of relative humidity, some chorionic alveoli containing electron dense structures. They presume that ions and/or molecules migrate during prehydration from inside the cyst to the alveolar layer.

Bioavailability of oxygen for the saline-water invertebrate <i>Artemia franciscana</i> was studied, since both oxygen concentration and oxygen diffusion rate change with salinity. Total haemoglobin concentration and the relative contribution of each of three haemoglobins was measured in specimens acclimated to different salinities and oxygen concentrations. Both haemoglobin concentration and contribution were influenced by salinity and the group observed (males, females with and females without eggs). 226 methanol and water extracts representing 74 mainly native plant species found in Amazonas State, Brazil, were tested at a standard concentration of 500 μg/mL for lethality towards larvae of the brine shrimp species Artemia franciscana. Several cytotoxic plant species were identified in this work: Aspidosperma marcgravianum, A. nitidum, Croton cajucara, Citrus limetta, Geissospermum argenteum, Minquartia guianensis, Piper aduncum, P. amapense, P. capitarianum, P. tuberculatum and Protium aracouchini. The results were analyzed within the context of the available traditional knowledge and uses for these plants.

Solid-phase extraction of ascorbic acid 2-sulfate from cysts of the brine shrimp <i>Artemia fransiscana</i> OMA

Two producers are described for the solid-phase extraction of ascorbic acid 2-sulfate from cysts of the brine shrimp <i>Artemia</i> . The first one is based on reversed-phase ion pair extraction on an octadecylsilica cartridge using tetrabutylammonium as the counterion, followed by elution with methanol. In the second procedure ascorbic acid 2-sulfate is retained on a DEAE silica cartridge and eluted with 0.1M sodium salicylate. Both approaches were used as part of a quantitative reversed-phase ion pair liquid chromatographic method with detection at 254nm. The brine shrimp <i><i>Artemia</i> franciscana</i> is widely used in aquaculture and basic research in areas ranging from molecular biology to evolution and ecology. A key of its life history involves the production of encysted embryos whose development is halted (enters diapause) at the gastrula stage.
